Following are the simplified procedures that we followed for the lab ... NOTE - I have not recorded time frames for you.
Procedure:
Procedure:
- Preserve in 10% formalin solution.
- Dehydrate in 95% ethanol.
- Place specimens in 30% acetic acid/70% ethanol/alcian blue stain.
- Place specimens in 15% hydrogen peroxide / 85% potassium hydroxide solution until depigmented.
- Put the specimen in 30% sodium borate solution, adding trypsin.
- Add alizarin red dye to 1% potassium hydroxide solution creating a deep reddish purple.
- Return specimen back into trypsin solution. NOTE - this step yeilded NO results. This step and the subsequents steps were as per https://www.researchgate.net/publication/280494794_The_Compleat_Guide_to_Clearing_and_Staining
- Store specimen in glycerin.
